Tamworth mum’s advice to any woman trying to conceive

A TAMWORTH woman who feared she would never have children because of a medical condition has come forward to tell her ‘story with a happy ending’, in a bid to give hope to other women.

Amanda and Mark Taylor are proud parents to five-month-old Lucas following IVF treatment.

Amanda (38) spent years suffering the painful and debilitating condition of endometriosis, before the birth of Lucas in October, following IVF treatment at Midland Fertility in Aldridge.

She told the Herald: “I’d like to say to other women suffering with endometriosis, don’t suffer in silence, speak to your GP to ensure you are getting the best advice possible for living with this illness, especially if you want to have a family at some point – it may be essential to act sooner rather than later.

“For those with endometriosis and currently trying to conceive, I would say don’t give up hope and if it’s not happening naturally then speak to your GP about your options before it’s too late.”

Endometriosis affects over 1.5 million women in the UK.

The condition means that cells like the ones that line the uterus grow elsewhere in the body.

These cells respond to a monthly cycle and cause pain, inflammation and scar tissue and make conception very difficult.

Amanda was referred for fertility treatment in March 2012 after trying to conceive for six years and following a history of recurring endometrial cysts, for which she had undergone numerous surgical procedures including a partial removal of her left ovary and the complete removal of one of her fallopian tubes.

After a failed first cycle of treatment Amanda underwent further surgery to remove yet more endometriotic cysts.

Following her recovery Amanda and Mark began a second treatment cycle in November 2012 which resulted in the positive pregnancy test that had eluded them so far.

Amanda said: “It had been such a long journey. I never thought I would ever be lucky enough to have a family. Since Lucas has arrived I feel blessed every day.”

“Endometriosis manifests itself in a variety of ways so diagnosis can be difficult and often delayed,” said Dr Abey Eapen, clinical lead at Midland Fertility.

“Recent research shows that there is now an average of seven and a half years between a woman first seeing a GP about her symptoms and receiving a firm diagnosis.

“Early recognition of the symptoms is vital so women can get a referral for fertility treatment if necessary.

“And there’s good evidence that pregnancy following IVF can ‘cure’ endometriosis so that the woman can conceive naturally again in the future.

“We’re delighted for Amanda and Mark about the birth of Lucas and are sure that her story will bring hope to other women who are living with endometriosis.”

The secret of fertile sperm

To better understand the causes of male infertility, a team of Bay Area researchers has been exploring the factors, both physiological and biochemical, that differentiate fertile sperm from infertile sperm. At the 58th Annual Biophysical Society Meeting, which took place Feb. 15-19, 2014, in San Francisco, Calif., the team presented its work to identify and characterize proteins known as ion channels, which are crucial for sperm fertility and expressed within a sperm cell’s plasma membrane.

“Any knowledge gained in this area may help create much-needed diagnostic testing and treatments for male infertility, which is in essence an idiopathic disease, because at this time 80 percent of male infertility cases can’t be diagnosed or treated,” said Melissa Miller, a postdoctoral fellow who will present the team’s findings at the meeting. Miller works in the labs of both of her co-authors, Polina Lishko of the University of California, Berkeley (UCB), and Yuriy Kirichok at the University of California, San Francisco (UCSF).

To study ion channels, the researchers are recording the electrical activity of sperm ion channels under strictly controlled conditions.

“Our labs have characterized three ion channels responsible for regulating calcium (CatSper), potassium (Slo1), and protons (Hv1) within sperm cells,” Miller said. “So far, the most well-studied is the sperm cation channel CatSper, which is exclusively expressed within sperm cells and represents an ideal target for development of a unisex contraceptive; no other cell in the body is known to express this protein.”

“We recently reported that the female hormone progesterone activates CatSper via a non-genomic pathway. In normal fertile spermatozoa, CatSper activity was greatly increased by the addition of progesterone. However, patch clamp recording from patient-derived sperm cells with CatSper deletion showed no response to progesterone nor did they produced basal CatSper current,” Miller said. “This shows direct regulation of the CatSper channel in human sperm by steroid hormones.”

Steroid hormones control the fundamental physiological functions via both traditional genomic and unconventional, nongenomic pathways. Previous research established that nongenomic signaling plays a vital role in both human sperm cell activation and pain perception modulation within neurons of the spinal cord. Because the molecular determinants of this pathway are poorly understood, Miller’s team is working to uncover the precise molecular signaling cascade that occurs in sperm cells and neurons upon progesterone treatment.

“The lack of CatSper activity is strongly correlated with male infertility, so identification of this endogenous signaling molecule would give us a novel biomarker for male fertility that could be immediately used in the clinic as a way to quickly assess sperm fertilization potential,” Miller said.

Overall, the knowledge gained from the team’s studies may be used in the future development of new pain management therapies, as well as diagnostic tests and treatments for male infertility.

Positive CB digital Ovulation test.

Very over joyed to to say I’ve got my BFP! Didn’t come on and announce on here straight away because it just didn’t seem real. Period day came and went and am now feeling very sick.

6 out of 7 of my friends all used conceive plus and fell pregnant

6 out of 7 of my friends all used conceive plus and fell pregnant within two months and the two friends who used pre seed fell one fell preg within a month and the other is still trying after 4.
